Is six days enough to see both West and East Sumba without rushing?

Yes, provided you cross once and only once. Three nights in the west cover Wanokaka, Praijing, Lapopu and a full Kodi day; the single Day 4 crossing takes indicatively under four hours; two east nights fit Walakiri, Prailiu and Rende. Anything shorter forces you to drop Kodi or Rende.

Can the 6D5N tour run east to west instead?

Yes. Fly into Waingapu on the TransNusa direct service, take the ikat villages and Walakiri first, cross to the west on Day 3, and finish with three lodge nights and a Tambolaka departure.
